The only professional antigen-presenting cells in the human epidermis are Langerhans (LC). They function as first line of defense against pathogens but also crucial for maintenance tissue homeostasis. One most well-known endocannabinoids, anandamide (arachidonoyl ethanolamide, AEA), is characterized by a general anti-inflammatory effect, it reduces production pro-inflammatory cytokines many immune cells, strength humoral and cellular response. In our current research, we investigated effect AEA regulating responses monocyte-derived LCs monitoring changes their maturation markers, viability, differentiation, cytokine production, naïve T cell activation capacity. First, to exclude possibility necrotic processes, performed viability tests using 7-ADD, which demonstrated that did not cause any death even when applied at high concentrations moLCs. Moreover, could demonstrate mostly induced concentrations, whereas TLR3-7/8 agonist-induced was affected.
